About Low-Pressure Liquid Chromatography
Low-pressure liquid chromatography or LPLC is an analytical technique that works with low pressure to drive the mobile phase of a column holding a stationary phase to separate mixtures of compound, which usually is complex due to differential partitioning. More often than not, chromatographic systems are often defined by characteristics such as the pressure. Hence, common acronyms would include LP (low pressure), and HPLC (high pressure liquid chromatography). In any case, for pressures <50 psi (~3 bar), this is called the low-pressure chromatography system. Low pressure is used when performing simple protein separation that does not require high resolution. The scale of low-pressure systems can start from simple right up to near semi-automated systems using gradient capabilities detectors, valves and fraction collectors among others. LPLC is based on the principle of differential partitioning of components in a mixture between a mobile phase (solvent) and a stationary phase (packing material). The stationary phase is usually made up of porous particles of silica, alumina, or polymer resins packed into a column. The mobile phase is pumped through the column, and the sample mixture is injected into the column. The sample components, as they pass through the column, interact differently with the stationary phase based on their physical and chemical properties, thus separating them.
