About Polymerase Chain Reaction
The polymerase chain reaction (PCR) is one of such laboratory techniques in great use for the multiplication of millions to billions of copies of a given DNA sample, allowing scientist to amplify very small amounts of DNA for in-depth study. PCR is most fundamental to genetic testing, research, and forensic science. It was invented by the American biochemist Kary Mullis in. PCR relies fundamentally on thermal cycling-consecutive cycles of heating and cooling are performed to allow DNA melting and enzyme-mediated DNA replication. In the process, primers, short single-stranded DNA fragments complementary to the target DNA region, are attached to the template; it uses a thermostable DNA polymerase. The PCR encompasses three crucial steps-denaturation, annealing, and extension. Denaturation occurs when the DNA double helix separates due to a high temperature. The temperature is then lowered during annealing, allowing the primers to bind to complementary DNA sequences. Finally, during extension, the DNA polymerase synthesizes the new DNA strand complementary to the template strand.
